Today I replaced a coil that had a broken spring in it. I have just recently replaced all of the coil boots but I have had a mis fire under throttle. I replaced the coil and put dielectric grease at the end of each coil boot. Now it is worse.

So before I go replacing stuff, could too much dielectric grease be the problem?

I wouldn't think so, every connector I could find on my car has gobs of dielectric grease packed in !

How many miles on the plugs, what make/model are they, did you pull and inspect them?

I use it liberally on connectors, never an issue...usually when this comes up, its a bad cop or more than one bad cop. Rarely an o2 sensor...but since you had a broken spring, that coil pack was most likely having to arc to fire, which will ruin one fast. This is why boots need to be changed more often....unfortunately if you wait until you have a misfire to replace them, you've already damaged atleast one of the packs internally. How long since your last plug change?

YES, YES IT CAN, AND DID. I will say today, on my 2018 Honda Accord I replaced the spark plugs for they had 90k miles on them. I put a small dab of dielectric grease onside every boot. When I started my engine holy crap, Misfiring, My braking control systems, stability control system lights "ALL" were lit up on my dash. What the hell just happened. I asked my Honda dealership could engine misfires cause all of my fault lights, he said absolutely for they are all tied into one another in one way or another. I took my coil packs out and sprayed with carburetor cleaner, ran micro fiber cloth down the tubes to remove all I could which is hard for even carburetor cleaner doesn't dissolve dielectric grease. I drove my car again, stopped my car and shut off then restarted, Low and behold "ALL" dash fault lights went out. I am confused as much as you guys. I used this on every connector, boot, Spark plugs my hole life and never had this happen. All I can say it did and WTF.
