Looking to upgrade a stock 5 speed V6 to around 300hp. I know gears will give it a good kick in the ass but Im looking for rwhp. Whats the best most cost effective hp for these motors with quality parts.

supercharger or turbo or nitrous to get 300hp+...bolt ons wont get you there

sorry h/c/i neither..best thing to do if you dont want to waste on a blower due to funds bolt ons will make your pony fun as hell..and you can get a bottle of zex 75 shot hp may not be in the 300hp but torque will be 350tq+ and torque is what wins races.

ok. what about h/c/i ? would that put me anywhere close?
SuperSix has numbers on h/c/i, but its only for Stage 2, in their words:

"4.0 SOHC Mustang, automatic transmission, single exhaust, Stage 2+ cylinder heads, gained 25 peak RWHP and 19 ft lbs RWTQ with non peak gains of up to 50 RWHP and 80 ft lbs RWTQ with no tuning changes on a Mustang chassis dyno".

Now with tuning you'll see an increase, and they have Stage 3 heads, but either way you probably won't hit 300 HP with H/C/I, but it certainly will give the car a kick in the pants thats there all the time unlike NOS.
